"What is it
Nimodipine is a calcium channel blocker used to prevent vasospasm secondary to subarachnoid hemorrhage (a form of cerebral hemorrhage). It was originally developed for the treatment of hypertension.

History
Nimodipine was patented in 1971 and approved for medical use in the United States in 1988.
It was approved for medical use in Germany in 1985.

Properties of Nimodipine

Indications
Prophylaxis and treatment of ischemic neurological deficits due to cerebral vasospasm after subarachnoid hemorrhage of aneurysmal origin.

Treatment of functional cerebro-organic changes resulting from aging, characterized by severe symptoms such as memory disorders, reduced mental capacity and concentration and emotional lability.

Before starting treatment with Nimodipine, it must be strictly determined that the symptoms are not caused by an underlying disease, requiring specific treatment.

Mechanism of Action
Nimodipine preferentially exerts an antivasoconstrictive and anti-ischemic cerebral action.

Nimodipine inhibits or eliminates vasoconstriction induced in vitro by different vasoactive substances (e.g. serotonin, prostaglandins, histamine) or by blood and its degradation products.

Nimodipine also has neuropharmacological and psychopharmacological properties.

Studies carried out in patients with acute changes in cerebral blood flow revealed that nimodipine dilates cerebral blood vessels and improves cerebral blood flow.

The increase in perfusion in already injured and insufficiently irrigated brain areas is generally more pronounced than in unaffected areas.

In patients affected by subarachnoid hemorrhage, nimodipine significantly reduces ischemic neurological deficits and mortality rate.

Nimodipine protects neurons and stabilizes their function, favors cerebral blood flow and increases tolerance to ischemia through actions on neuron receptors and on cerebrovascular receptors linked to calcium channels.

Other studies demonstrated that such properties did not cause theft effect.

At a clinical level, nimodipine has been shown to improve memory and concentration disorders in patients with functional cerebro-organic changes resulting from aging.

Other typical symptoms were also favorably influenced.

These findings were demonstrated through the assessment of the global clinical picture, the diagnosis of individual disorders, the observation of behavior and performance evidenced in psychometric tests."
