Gas chromatography-mass spectrometry (GC-MS) is probably the most accurate; but, it’s labor intensive, so it doesn’t seem to be offered much anymore. My conclusion based on poking around in the literature (when I had the same question about techniques) was that LC-MS-MS is more than accurate enough for ranges commonly encountered in males. It might be a little iffy for low range females.

If you are getting these tests to figure out what’s up with your large pet rat then I would pair the testosterone test with a few others to get a better picture of your large rats endocrine system. Whatever test technique you settle on/is available consider:

Total testosterone
Free testosterone
LH
FSH
Total estrogen
SHBG
Prolactin
Vitamin D


If you think that your large rat is having low testosterone symptoms and want to establish possible causes (or alternative causes) then thyroid tests can be useful as leydig cells are dependent on T3 (among other things) and low thyroid function seems to mimic certain low testosterone symptoms. So, consider adding:

TSH
T4
T3
