Vitamin B-6 and colorectal cancer risk: a prospective population-based study using 3 distinct plasma markers of vitamin B-6 status
Background: Higher plasma concentrations of the vitamin B-6 marker pyridoxal 5′-phosphate (PLP) have been associated with reduced colorectal cancer (CRC) risk. Inflammatory processes, including vitamin B-6 catabolism, could explain such findings.

Objective: We investigated 3 biomarkers of vitamin B-6 status in relation to CRC risk.

Design: This was a prospective case-control study of 613 CRC cases and 1190 matched controls nested within the Northern Sweden Health and Disease Study (n= 114,679). Participants were followed from 1985 to 2009, and the median follow-up from baseline to CRC diagnosis was 8.2 y. PLP, pyridoxal, pyridoxic acid (PA), 3-hydroxykynurenine, and xanthurenic acids (XAs) were measured in plasma with the use of liquid chromatography–tandem mass spectrometry. We calculated relative and absolute risks of CRC for PLP and the ratios 3-hydroxykynurenine:XA (HK:XA), an inverse marker of functional vitamin B-6 status, and PA:(PLP + pyridoxal) (PAr), a marker of inflammation and oxidative stress and an inverse marker of vitamin B-6 status.

Results: Plasma PLP concentrations were associated with a reduced CRC risk for the third compared with the first quartile and for PLP sufficiency compared with deficiency [OR: 0.60 (95% CI: 0.44, 0.81) and OR: 0.55 (95% CI: 0.37, 0.81), respectively]. HK:XA and PAr were both associated with increased CRC risk [OR: 1.48 (95% CI: 1.08, 2.02) and OR: 1.50 (95% CI: 1.10, 2.04), respectively] for the fourth compared with the first quartile. For HK:XA and PAr, the findings were mainly observed in study participants with <10.5 y of follow-up between sampling and diagnosis.

Conclusions: Vitamin B-6 deficiency as measured by plasma PLP is associated with a clear increase in CRC risk. Furthermore, our analyses of novel markers of functional vitamin B-6 status and vitamin B-6–associated oxidative stress and inflammation suggest a role in tumor progression rather than initiation.

And here is perhaps a very interesting study on the topic. Isn't it cool that here in the US so many of us must eat government-mandated "fortified" foods containing folic acid, non-heme iron etc.?

Cancer Incidence and Mortality After Treatment With Folic Acid and Vitamin B12
Context Recently, concern has been raised about the safety of folic acid, particularly in relation to cancer risk.

Objective To evaluate effects of treatment with B vitamins on cancer outcomes and all-cause mortality in 2 randomized controlled trials.

Design, Setting, and Participants Combined analysis and extended follow-up of participants from 2 randomized, double-blind, placebo-controlled clinical trials (Norwegian Vitamin Trial and Western Norway B Vitamin Intervention Trial). A total of 6837 patients with ischemic heart disease were treated with B vitamins or placebo between 1998 and 2005, and were followed up through December 31, 2007.

Interventions Oral treatment with folic acid (0.8 mg/d) plus vitamin B12 (0.4 mg/d) and vitamin B6 (40 mg/d) (n = 1708); folic acid (0.8 mg/d) plus vitamin B12 (0.4 mg/d) (n = 1703); vitamin B6 alone (40 mg/d) (n = 1705); or placebo (n = 1721).

Main Outcome Measures Cancer incidence, cancer mortality, and all-cause mortality.

Results During study treatment, median serum folate concentration increased more than 6-fold among participants given folic acid. After a median 39 months of treatment and an additional 38 months of posttrial observational follow-up, 341 participants (10.0%) who received folic acid plus vitamin B12 vs 288 participants (8.4%) who did not receive such treatment were diagnosed with cancer (hazard ratio

A total of 136 (4.0%) who received folic acid plus vitamin B12 vs 100 (2.9%) who did not receive such treatment died from cancer (HR, 1.38; 95% CI, 1.07-1.79; P = .01). A total of 548 patients (16.1%) who received folic acid plus vitamin B12 vs 473 (13.8%) who did not receive such treatment died from any cause (HR, 1.18; 95% CI, 1.04-1.33; P = .01). Results were mainly driven by increased lung cancer incidence in participants who received folic acid plus vitamin B12. Vitamin B6 treatment was not associated with any significant effects.

Conclusion Treatment with folic acid plus vitamin B12 was associated with increased cancer outcomes and all-cause mortality in patients with ischemic heart disease in Norway, where there is no folic acid fortification of foods.

Fear mongering. If you scroll down it even says their results did not prove b vitamins cause lung cancer and other studies with high dose b vitamins did not have the same results. This is the problem with using correlations in science, journalists tend to jump on the results without understanding the limits of the methodology. By the time it gets to us, we get an abstract or a summary at best with out the full study to analyze. As a rule of thumb, I assume most research is bogus because most of it is. The best research is hard science and uses animals in a randomized controlled lab experiment. Soft sciences use correlations to try and fit a predefined conclusion.

B12 does, which is also obscured by the misleading title.

That is mostly because people take several thousand times daily doses.
Also, B12 is a micronutrient, and also not a vitamin because it is manufactured inside the body; that is why it does not exist in nature in plants, soil, roots, etc and can only be sourced from animals and their products. It is a complex enzyme, with cobalt inside. No idea who decided to call it B vitamin and add it to the complex, thus ruining one of the otherwise most beneficial things people can supplement.

Seems biotin rise insulin resistance? Can supplementation in high doses boost stress response?

I stay away from all B vitamins. All. The best way to get vitamins is thru natural food source.
