Niacin, the fountain of youth
”Ristow and his team's work now suggests that the activity of sirtuins actually prolongs life in roundworms. According to the study, however, the life-prolonging effect is not down to gene regulation, as has often been supposed in the past. Instead, the effect is due to the conversion of niacin into nicotinamide. Studying genetically modified roundworms that were unable to convert nicotinamide into certain other metabolic products, the scientists did not observe any lifespan extension, even after overexpression of sirtuins, which otherwise lead to an increased life expectancy.”
So if we can compare extending life in worms with extending life in humans by using niacin/niacinamide. It seems that niacinamide is what is needed to extend lifespan likely through boostung NAD+. Thus using nicotinamide would be beneficial. Another way would be taking niacin and exercising as exercise increases sirtuins thus converting more niacin to nicotinamide. Niacin by itself also increase sirtuin expression which makes sense as the body likely sense increased niacin levels thys upregulating the conversion.
Sirtuins are also on wikipedia and other website promoted as the main factor for positive effects of exercise. So sirtuin activators could be called exercise mimetics
