It seems that pregnenolone is finally starting to look exciting to the medical industry. In addition for its intensive studies for a variety of mental issues, now studies are coming out demonstrating its endotoxin (LPS) antagonism effects. As most forum users know, endotoxin exerts its effects mainly through activation of the TLR4 receptor, and pregnenolone prevented proper activation of TLR4 by endotoxin. The article states that this TLR4 blocking effect of pregnenolone may turn it into promising treatments for alcoholism, schizophrenia, epilepsy, MS, sepsis, PTSD, Alzheimer, CVD, etc - i.e. all conditions associated with abnormal TLR4 activation. The TLR4 antagonism may also explain the benefits of pregnenolone for rheumatoid arthritis (RA) as demonstrated by multiple human studies back in the first half of the 20th century.
The study showed that both pregnenolone and its downstream metabolite allopregnanolone prevented the inflammatory effects associated with TLR4 activation. Allopregnanolone was slightly more potent that pregnenolone but that difference can be overcome by simply increasing the pregnenolone dose. Since the potent anti-inflammatory effects were observed a concentration of 1 uM/L this suggests human doses of 30mg-50mg should be enough to replicate the study design. Also, while this study did not look at progesterone there are a number of other ones that have found progesterone is also a functional TLR4 antagonist in concentrations even lower then 1 uM/L.

Would topical application have the same effects on endotoxin, or would it have to be oral so it gets to the intestine?

Topical would get metabolized extensively by the skin, and it may actually result in both allopregnanolone and pregnenolone entering the blood.

Thanks, so I'm guessing topical would have the same effect but likely in a smaller dose then with the oral.

Thanks, so I'm guessing topical would have the same effect but likely in a smaller dose then with the oral.

That I don't know, as it would depend on the solvent and how much gets absorbed/metabolized. But the experience from topical seems quite different from oral for most people who tried both routes.

Pregnenolone seems to give me acne. Estrogenic impurities? I wonder if any of the pregnenolone is actually pure.

More likely it is getting extensively metabolized into downstream steroids like DHEA and androstenedione, and then possibly estrogen. An old study (and the only one I have been able to find) with topical pregnenolone found it elevated DHEA and androstenedione in a dose of just 5mg topically daily. The study does not have full text so I cannot see what the solvent was, but the DHEA/androstenedione elevations an easily explain the acne.

Anecdotally I get acne with 25mg sublingual tabs, but zero negative sides with pansterone in dmso applied to scrotum in a single bottle dose. Actually stokes libido for me.

Also, pardon my ignorance, but why can preg be sold no problem as an over-the-counter supplement but allopreg can only now be acquired because Brex got FDA approval? Are not these two molecules similar enough that they'd be classified in the same way? thanks

Search the forum for "pyridoxamine" (no quotes) and you will see just how evil FDA can be.
