NMDA / Schizophrenia

Current research in the psychiatric field suggests that probably the primary cause of schizophrenia and schizoaffective disorders is in NMDA receptors. In particular, a hypofunction has been noted, therefore glutamate probably cannot enter the canals ... consequently energy production (krebs cycle) is compromised as well as many other functions.

Pregnenolone was seen as a positive modulator of NMDA receptors and also in GABAa.

The same thing the serine being NMDA agonist.

Has anyone had any experiences in this particular case?

All I know is when NMDA gets blocked too much by Magnesium or Zinc, I get a bit spacey and depersonalization ensues. I’ve also taken pregnenalone and had that occur so who the hell knows

Sarcosine works for the negative symptoms of schizophrenia by allowing the NMDA channels to open up

in that frame of mind, can you socialise ? or can't you be bothered.

I become like a mute when I take pregnenolone.

One hundred percent cannot socialize if NMDA is blocked too much by Magnesium, I just feel out of synchronization with environmental cues, but otherwise relaxed and blah. I could barely think or hold a conversation.

Pregnenalone sometimes did this as well at low doses, but who knows what mechanism is behind it

Seems to be a sweet spot with Pregnenalone whereby just the right amount makes me Social

Yea, sarcosine (N-methylglycine). There's also aniracetam (+ noopept + alpha-GPC). Even 1g flushing niacin + 1g vitamin C 3x/day works for some.

Another theory suggests there's an autoimmune attack on NMDA receptors as well, implying megadose vitamin D3.

what about the high DAO inhibition?
D-amminoacido ossidasi? DAAO abbassa la D-serina. La glicina può anche fungere da coagonista (con glutammato) per attivare i recettori NMDA. [/ QUOTE]

Yes DAAO is high so serine is consume fast i think. What is a good serine dosage ? and for DAAO inibhitor ?

Glycine is a coagonist like D-serine, hence sarcosine (N-methylglycine).

Unfortunately I don't find sarcosine in Europe but only D-Serine. 2-3gr daily is good ? cycling ? I have read that B1 and acetyl cysteine are DAO inhibitors and also cimetidine an H2 antihistamine.

Glycine work at a lot dosage (60gr) daily

Unsure about dose, but I suppose taking it directly may work. Yea, higher amounts of glycine would also work. The added methyl group in the other product lowered the amount needed.

I though schizophrenia was an overactivation of NMDA and elevated glutamate activity. Ive experienced a distorted perception of time which in common in schizos but i get real messed up from glycine. I rcm reading Jack Kruse' post on schizophrenia and alterations in internal clocks.

I have massive problems with deja vu and apathy and sleep problems.

Anthony Peake writes about alternate states and schizophrenia and it does have a "spiritual" element.

Pregnenolone gave me selective mutism and delusional telepathic thinking sometimes.
There's also a "study" saying schizphrenics have different head shapes

Why is deja vu a problem? By the way mucuna pruriens gives pll deja vu and is a dopamin precursor. What do you think about the dopamin theory of schizp? Have you treid red light therapy?
