...and what the drug does is , simply blocking the integrated stress response !
This article is a good read . The researchers seem pretty legit , although I'm sure they dont know about RP ,but they do look at inflammation and cellular health as important parts in chronic diseases .
Pretty telling that this kind of research normally wouldn't have happened ,if it wasn't for a family that supported them financially (The Rogers Family Foundation)

Plus: As mentioned in the article ISRIB also treats "aggressive prostate cancer " ,which suggests a very systemic mechanism of action.

One might think that interfering with the ISR, a critical cellular safety mechanism, would be sure to have serious side effects, but so far in all their studies, the researchers have observed none. This is likely due to two factors, Walter says. First, it takes just a few doses of ISRIB to reset unhealthy, chronic ISR activation back to a healthier state, after which it can still respond normally to problems in individual cells. Second, ISRIB has virtually no effect when applied to cells actively employing the ISR in its most powerful form – against an aggressive viral infection, for example.

Naturally, both of these factors make the molecule much less likely to have negative side effects – and more attractive as a potential therapeutic. “It almost seems too good to be true, but with ISRIB we seem to have hit a sweet spot for manipulating the ISR with an ideal therapeutic window,” Walter said.

Yup, and if it helps with mental decline /alzheimers it might just as well help stopping and partially reverse aging itself.

Yes and no.

The ISR is a homeostatic response to different stressors. Ideally it restores/helps to restore health/function within the cell or it helps to bring about apoptosis.

That are basically the two poles.
In aging or impairment of other causes it seems that neither poles can be reached sufficiently, so you either have inability to restore a homeostatic health or it causes apoptosis too quickly. Blocking ISB helps only one way thus and is not able to generaly alleviate aging. That was the no.
Because the inadequate ISR must have causes. And they are (certainly) immunological and probably metabolic/bionergetic

The article you linked towards the end mentions that ISRIB shows effects that show promise to also rejuvenate T-cells and/or other immunity mechanisms. If that’s the case this is indication that it influences more upstream factors that are impaired by alleviating downstream itself and helping to set in motion a positive reinforcement/loop. Then it might truly be a breakthrough towards longer healthspan.

Yes , i am certainly having high hopes with this one , so I'm looking forward to seeing more studies on it .

At Sigma, it costs 100 USD for 5mg.
Oh wow, I didn't know it was already available .

I emailed it to him .
He says he thinks pregnenolone can do the same but with less side effects .
